Looking at biological samples with SEM is tricky:
- It has to be done in vacuum, which will cause water to evaporate and some structures will explode, deflate or deform.
- The electron-beam induces chemical reactions like polymerization. Small biological features wiggle around while they are more and more deformed.
- Organic structures have low electrical conductivity and are prone to getting electrically charged. This causes imaging problems, e.g. making images fuzzy over time.
