I found ignore interferes with searching DU for posts you know you've seen. Also, I think a post by someone who you do not have on ignore replies to someone who is on ignore gets hidden alone with the post by the person you did put on ignore.

Still, you can't beat ignore for a more pleasant DU.

No, I don't count them by hand! I paste the contents of my ignore list in a text editor (e.g. Notepad++) and get the # of lines of text. Pretty hard to do this on my phone, so I don't have the exact # right now.
