There is a setting that people with many followers generally use which notifies them when only people THEY follow reply or like their tweet, rather than the default setting of notifying when any rando replies/likes.

Nevertheless, everyone else lurking can see your replies, so it does shape the conversation. But it’s less satisfying than being able to get in their face with your reply.
