Why? He's already said publicly that the commercial is BS and quotes him out of context.

If Donnie had an ounce of shame, that sort of thing might work. But Donnie is completely shameless, and he'd just replace Fauci with someone more personally loyal and more incompetent.
Authoritarians purge those with any integrity and replace them with loyalists instead. In that environment, protest resignations actually help the authoritarian solidify control.

Trump can't and/or won't fire Fauci.
Better to poke Trump in the eye by defying him by exposing his lies AND out-lasting him.
I don't worship at the altar of Fauci by any means, but the better victory is to flip him the bird and still be there after he's gone.
